DNA-based forensics is largely centered around PCR-based amplification of a few dozen STR markers. While this assay is powerful, it does not work for samples with small amounts of fragmented DNA like rootless hair shafts. My lab and company have pioneered high-throughput sequencing technologies that enable new approaches in DNA bases forensics. These include forensic genetic genealogy from difficult samples and non-PCR based tests for identity. These technologies have provided investigative leads and probative evidence in some of the highest priority cold cases in the United States.
